The degree of efficiency of labeling can be significantly improved by (1) using a tRNA-mixed solution which comprises substantially only stretched tRNA having initiation tRNA removed therefrom and utilizing a synthesis system (an in vitro transcription/translation system) which does not contain methyonyl tRNA transformylase and 10-formyl-5,6,7,8-tetrahydrofolic acid; (2) isolating/purifying labeled methyonyl initiation tRNA; and/or (3) controlling the time required for the labeled-protein synthesis reaction.

Le degré d'efficacité du marquage peut être considérablement amélioré (1) en utilisant une solution d'ARNt mélangés qui ne comprend pratiquement que des ARNt étirés dont l'ARNt d'initiation a été éliminé et en utilisant un système de synthèse (système de transcription/traduction in vitro) qui ne contient pas de méthyonyl ARNt-transformylase ni d'acide 10-formyl-5,6,7,8-tétrahydrofolique ; (2) en isolant/purifiant l'ARNt d'initiation du méthyonyle marqué ; et/ou (3) en réglant le temps requis pour la réaction de synthèse de la protéine marquée.

本願発明者らは、(1)開始tRNA を除いた実質的に伸長tRNAのみからなるtRNA混合液を用い,さらにメチオニルtRNAトランスフォルミラーゼ,10−フォルミル5,6,7,8-テトラヒドロ葉酸を含まない合成系(in vitro転写・翻訳系)を利用する、(2)標識されたメチオニル開始tRNAを分離精製する、及び/又は(3)標識タンパク質合成反応時間も制御することによって、標識効率を大幅に改善した。
