本発明では、紫外線又は発色試薬に対する光学応答性を有する試料に対して、試料と同じ光学応答性を有する第一の分離剤層と、これに隣接して併設されている、前記光学応答性とは異なる光学応答性を有する第二の分離剤層とを有する薄層クロマトグラフィープレートを用い、第一の分離剤層に試料を展開させた後に、第一の分離剤層から第二分離剤層に向けてスポットをさらに展開させ、第二の分離剤層において紫外線の照射又は発色試薬の発色処理によってスポットを検出する。
