The present invention relates to: a method for manufacturing a nano-liposome containing a nucleic acid without an extrusion process, comprising the step of mixing a lipid solution in which a lipid is dissolved and an aqueous solution in which a nucleic acid is dissolved into a mixed solvent containing diethyl ether and ethanol; and a kit for manufacturing a nano-liposome comprising the mixed solvent and the lipid mixture.
