The present invention relates to a siRNA molecule having improved specificity for inhibiting target gene expression and a decreased off-target effect, the siRNA molecule having a structure of asymmetric 3'-overhangs and consisting of sense and antisense strands, wherein one of the two strands has 3'-overhangs consisting of DNA of 1 to 3 nucleotides and the other has 3'-overhangs consisting of RNA of 1 to 3 nucleotides.

본 발명은 목적 유전자의 발현 억제 특이성이 증가되고 오프-타겟 효과가 감소된 siRNA 분자로서, 상기 siRNA 분자는 센스 및 안티센스 가닥으로 이루어져 있으며, 상기 두 가닥 중 하나의 가닥은 1 내지 3개의 뉴클레오티드의 DNA로 구성된 3'-오버행을 가지며, 나머지 하나의 가닥은 1 내지 3개의 뉴클레오티드의 RNA로 구성된 3'-오버행을 가지는 것을 특징으로 하는, 3'-오버행이 비대칭적인 구조를 갖는 siRNA 분자에 관한 것이다.
