The method includes obtaining nucleic acid molecules encoding heavy and light immunoglobulin chains (or variable regions thereof) of multiple immunoglobulins from an animal; obtaining mass spectra information of peptide fragments of a population of polyclonal immunoglobulins that specifically bind to an antigen of choice; comparing and/or correlating the mass spectra information of the peptide fragments of the polyclonal immunoglobulins with predicted mass spectra information of predicted amino acid sequences encoded by the nucleic acid molecules, and then assembling the heavy and light chains to create an antibody (or variable region thereof) that specifically binds to the antigen.

Le procédé comprend l'obtention de molécules d'acide nucléique codant pour des chaînes lourdes et légères d'immunoglobuline (ou des régions variables de celles-ci) d'immunoglobulines multiples d'un animal; l'obtention d'informations de spectre de masse de fragments peptidiques d'une population d'immunoglobulines polyclonales qui se lient spécifiquement à un antigène de choix; la comparaison et/ou la corrélation des informations de spectre de masse des fragments peptidiques des immunoglobulines polyclonales avec des informations de spectre de masse prédites de séquences d'acides aminés prédites codées par les molécules d'acide nucléique, puis l'assemblage des chaînes lourdes et légères pour créer un anticorps (ou une région variable de celui-ci) qui se lie spécifiquement à l'antigène.
